Abstract

The preparative expression of genes is effected in a cell-free system of conjugated transcription/translation comprising, as substrates, amino acids, ATP, GTP, CTP and UTP, the genes being used in the form of DNA molecules. As a result, products of gene expression are obtained in the system, including AMP, ADP, GDP, CDP, UDP, pyrophosphate and inorganic phosphate. During the functioning of the cell-free system of conjugated transcription/translation as the substrates are being used up with subsequent generation of the products, those products of gene expression which include polypeptides, AMP, ADP, GDP, CDP, UDP, pyrophosphate and inorganic phosphate are extracted from the system. Simultaneously substrates are introduced into the system in the form of amino acids, ATP, GTP, CTP, UTP for maintaining their initial concentration.
